ABSTRACT

The continuous growth of the global human population and the depletion of natural energy resources are posing a threat to the environment and the sustainable supply of food and energy. To address this issue, a solution known as "green technology" has been developed, focusing on the use of eco-friendly approaches. One of these approaches involves harnessing the power of cyanobacteria, which are ancient microorganisms that have existed on Earth for a long time. This approach was used to probe the diversity of phototrophic microorganisms in Wheat samples. The presence of nif genes allows cyanobacteria to x nitrogen which plays a vital role in nitrogen cycle and biological processes such as plant growth and soil fertility. Wheat production test in current study evaluates the yield and quality of wheat crops. It shows factors such as improvement in growth, heat resistance and environmental adaptability, improvement in production; provide insights for optimizing cultivation practices and ensuring successful high yield. Identication of Cyanobacteria for their combination of nif and molecular identity is prior for production of bio fertilizers. Cyanobacteria can be cultivated on a large scale to produce biomass that has multiple uses, including biofertilizer, secondary metabolites, cosmetics, and medicines. In the eld of agriculture, production of healthy crop cyanobacterial biofertilizer are being increasingly used in eco-friendly and sustainable practices. These biofertilizer are produced through mass cultivation of cyanobacteria and offer several benets.

Resumen Introducción: El embalse Cerrón Grande de 135 km2 es el humedal más grande de El Salvador, reconocido como "Humedal de Importancia Internacional" por la Convención Ramsar. Además, es uno de los ecosistemas más contaminados a nivel nacional, enfrentando serios problemas, entre ellos, las proliferaciones de cianobacterias. Objetivo: Evaluar las relaciones entre los factores ambientales y la distribución espacial y temporal de las cianobacterias en el embalse a lo largo de un año hidrológico. Métodos: Extrajimos agua con cianobacterias de seis sitios fijos, desde octubre 2018 hasta septiembre 2019. Resultados: Identificamos 16 géneros, el más abundante Microcystis sp. Además, Dolichospermum sp.; Microcistis sp. alcanzaron 1.5 x 106 células/ml en junio 2019, principalmente en el sector noroeste, que es poco profundo y cercano a las zonas de confluencia de afluentes altamente contaminados que llegan al embalse. El sector sureste más profundo tenía bajas concentraciones de estos organismos. Los factores ambientales mayormente relacionados con dicho comportamiento fueron: fósforo, la conductividad eléctrica y el valor de Secchi, como se esperaba de un cuerpo eutrófico. Conclusiones: La variación en la concentración celular de cianobacterias en este embalse está altamente influenciada por la lluvia, la mezcla de agua y el contenido de nutrientes.


Abstract Introduction: The 135 km2 Cerrón Grande reservoir is the largest wetland in El Salvador, recognized as "Wetland of International Importance" by the Ramsar Convention. It is also one of the most polluted ecosystems nationwide, facing serious problems, among them, a proliferation of cyanobacteria. Objective: To assess the relationships among environmental factors and the spatial and temporary distribution of cyanobacteria in the reservoir throughout a hydrological year. Methods: We extracted water with cyanobacteria from six fixed sites, from October 2018 through September 2019. Results: We identified 16 genera, the most abundant Microcystis sp. And Dolichospermum sp.; Microcystis sp. reached 1.5 x 106 cells/ml in June 2019, mainly in the north-west sector, which is shallow and close to the confluence zones where highly polluted tributaries reach the reservoir. The deeper south-east sector had low concentrations of these organisms. The environmental factors mostly related to this behavior were: phosphorus, electrical conductivity and Secchi value, as expected from a eutrophic body. Conclusions: Variation in the cellular concentration of cyanobacteria in this reservoir is highly influenced by rain, water mixing and nutrient content.

As florações são resultado de interações entre fatores físicos, químicos e biológicos, sendo caracterizadas pelo crescimento massivo de microrganismos. As proliferações de cianobactérias indicam a deterioração na qualidade da água e podem ser consideradas problemas de saúde pública, devido à capacidade de produção e liberação de toxinas pelas cianobactérias nos ambientes aquáticos, especialmente nos reservatórios de abastecimento. Assim, este trabalho teve como objetivo verificar, a partir de uma revisão de literatura, a ocorrência de florações de cianobactérias e cianotoxinas e os efeitos especialmente para saúde pública. A metodologia utilizada foi revisão narrativa de trabalhos selecionados seguindo os critérios de inclusão. Foi verificada a ocorrência de 74 espécies de cianobactérias, sendo Microcystis aeruginosa e Raphidiopsis raciborskii as predominantes. Espécies potencialmente tóxicas foram encontradas em todas as florações relatadas nos trabalhos realizados na região Nordeste. As florações de cianobactérias ocorreram principalmente em reservatórios da região Nordeste, juntamente com cianotoxinas (microcistinas, cilindrospermopiscina e saxitoxina). Têm sido frequentes florações com cianotoxinas em sistemas aquáticos, ocasionando alterações no ambiente aquático. Essas modificações alcançam diversos níveis tróficos e podem alcançar o homem, sendo problema de saúde pública.


The blooms are the result of interactions between physical, chemical, and biological factors, and are characterized by the explosive growth of microorganisms. The proliferations of cyanobacteria indicate deterioration in quality and can be considered public health problems due to the ability of cyanobacteria to produce and release toxins in aquatic environments, especially in supply reservoirs. Thus, this study aimed to review the literature to verify the occurrence of cyanobacteria and cyanotoxin blooms and the effects especially for public health. The methodology used was narrative review of selected papers according to the inclusion criteria. The occurrence of 74 cyanobacteria species was verified, with Microcystis aeruginosa and Raphidiopsis raciborskii being the predominant ones. Potentially toxic species were found in all blooms studies carried out in the Northeast region. Cyanobacterial blooms were found mainly in reservoirs in the Northeast region, along with cyanotoxins (microcystins, cylindrospermopsin, and saxitoxin). Blooms with cyanotoxins have been frequent in aquatic systems, causing changes in the aquatic environment. These changes reach several trophic levels, and can reach man, being a public health problem.


Las floraciones son el resultado de interacciones entre factores físicos, químicos y biológicos, caracterizadas por el crecimiento explosivo de microorganismos. La proliferación de cianobacterias indica un deterioro de la calidad y puede considerarse un problema de salud pública debido a la capacidad de las cianobacterias de producir y liberar toxinas en ambientes acuáticos, especialmente en los embalses de abastecimiento. Así, este estudio tenía como objetivo revisar la literatura para verificar la ocurrencia de floraciones de cianobacterias y cianotoxina, así como sus efectos especialmente en la salud pública. La metodología utilizada fue la revisión narrativa de los documentos seleccionados de acuerdo con los criterios de inclusión. Se verificó la aparición de 74 especies de cianobacterias, de las cuales Microcystis aeruginosa y Raphidiopsis raciborskii fueron las más predominantes. Se encontraron especies potencialmente tóxicas en todos los estudios de floración realizados en los estados de la región Nordeste de Brasil. Las floraciones de cianobacterias se encontraron principalmente en embalses en la región Nordeste, junto con cianotoxinas (microcistinas, cilindrospermopsinas y saxitoxinas). Ha habido frecuentes floraciones con cianotoxinas en los sistemas acuáticos, causando cambios en el medio acuático. Estos cambios alcanzan varios niveles tróficos y pueden llegar al hombre, siendo un problema de salud pública.

ABSTRACT

During the summer growing seasons of 2021 and 2022, two field experiments were conducted at Ismailia Agricultural Research Center Station(Latitude 30? 35? 41.901? N and Longitude 32? 16? 45. 843?E) to study the effect of cyanobacterial inoculation (Anabaena oryzae (A. oryzae) and Nostoc mascarum (N. mascarum)) on peanut yield, quality and certain soil biological activities under various nitrogen fertilization conditions and three types of applications, thefirst treatment was carried out as coating seeds with powder of individual of each cyanobacterial strain and the before planted, second treatment seeds were drenched withsuspension of eachcyanobacterial strain individually andthe last treatment was by foliar doses after 15, 45 and 60 days from seeds planting. Results showed that applying cyanobacteria inoculation to peanut plants generally enhanced peanut plant growth, leading to significantly higher yields of peanut and grains than uninoculated treatments. Treatment of N. mascarum + 75% N recorded the highest peanut yield and plant characteristics followed by N. mascarum+ 75% N in soil drench application compared to other tested treatments and types of applications. Cyanobacteria enhanced the amount of N, P, Kand Ca in peanut plants overall. By increasing the total chlorophyl, carotenoids, dehydrogenase, urease activities and nutrients in the peanut rhizosphere, cyanobacteria inoculation had a favorable impact on soil fertility. In general, cyanobacteria inoculation with 75% nitrogen amounts can benefit under peanut growth in sandy soil conditions.

ABSTRACT

Cyanobacteria are microorganisms found in different parts of the world. Some genera are cyanotoxins producers a sodium channel blockingneurotoxin (saxitoxins). Some homeopathic preparations have been identified as remedial action on toxicity models in Artemia salina. This study aimed to observe whether homeopathic products influence the toxicity ofR.raciborskiiextract onA.salinaby inducing cyst hatching arrest, anembryo bioresiliencemodel previously developed in our laboratory (Pinto et al., 2021; Mohammad et al., 2022). Thus, previous toxicity testswere carried out on cysts in 96-well plates, using different concentrations of the extract obtained from regular cultivation of R. raciborskii in HCl 0.05M, whose strain, named T3, is kept in the laboratory of Cyanobacteria at FURGS, Brazil.The standardization of toxin concentration was based on an established scale developed at FURGS, in which the number of T3 filaments is associated withspecific saxitoxin concentrationsdefined by chromatography. The concentration of 2.6 µg/L was chosen since it reducedthe cysthatching rate by 30%, the ideal level to observe embryo bioresilience. Then, a screeningstudy with 22 homeopathic preparations was tested blind in three experimental series, in duplicate,against threecontrols (unchallenged, water,and succussed water)for possible toxicity attenuationon Artemia salinacysts hatching rate. Homeopathic medicines were prepared in pure,sterile water from a stock homeopathic solution, one potency below the working potency. After the 1:100 dilution, 100 succussions were made using a robotic arm (Denise, Autic). The medicines were inserted into the seawater on a 10% basis. Due to the high sensitivity of A. salinato the circalunar variations, all experiments were performed during the first quarter moon. Statistical analysis was performed by two-way ANOVA followed by Tukey, with α=0.05. The most significant results indicative of bioresilience improvement were seen after the treatment with Nitric acidum6 cH, Plumbum metallicum6 cH, isotherapic 200 cH, and hydrochloric acid 1 cH being the last one used as a vehicle of the extracts. Thus, these preparations were chosen to be used in further experiments. In conclusion, the Artemia salinamodel has also beenuseful to study bioresilienceimprovement by homeopathic medicines after intoxication with saxitoxin.

RESUMO A escassez hídrica e o aumento da demanda de água para usos múltiplos incentivaram a construção de diversos reservatórios e sistemas de transferências hídricas entre bacias hidrográficas em diversas partes do mundo, principalmente em regiões áridas e semiáridas. Apesar de essas medidas mitigarem os aspectos quantitativos do abastecimento, a diminuição da qualidade da água disponível pode se tornar uma limitação importante. Este estudo teve por objetivo avaliar a qualidade da água no sistema de transferência que abastece a Região Metropolitana de Fortaleza, Ceará. Foram analisados 20 parâmetros de qualidade de água ao longo do sistema. Aplicou-se em seguida a análise de agrupamento utilizando a distância euclidiana associada ao método de ligação de Ward para agrupar os 13 pontos de coleta que foram estabelecidos conforme suas similaridades. A piora da qualidade da água ficou evidente ao longo do sistema, com o aumento da concentração de cianobactérias, pH, nitrogênio total, oxigênio dissolvido, turbidez e clorofila-a. Com relação às cianobactérias, duas espécies mostraram-se dominantes — Oscillatoria sp. e Microcystis sp. — e sete abundantes — Merismopedia sp., Cylindrospermopsis sp., Pseudanabaena sp., Anabaena sp., Aphanizomenon sp., Aphanocapsa sp. e Dolichospermum sp. A presença de cianobactérias potencialmente tóxicas e a piora da qualidade da água tornam a realização de monitoramentos frequentes e o desenvolvimento de sistemas de alerta essenciais para a adequada gestão dos recursos hídricos, tanto do ponto de vista ambiental quanto do de saúde pública.


ABSTRACT Water scarcity and increasing water demand for multiple uses encourage the construction of reservoirs and water transfer systems between watersheds in various parts of the world, especially in arid and semi-arid regions. Although these measures may mitigate the quantitative aspects of water supply, the quality decrease of the available water may become an important limitation. This study aimed to evaluate the water quality in the water transfer system that supplies the metropolitan region of Fortaleza, Ceará. Twenty water quality parameters were analyzed throughout the system followed by the application of the Euclidean distance and Ward's method to group the 13 sampling points according to their similarities. The worsening of the water quality was evident with increasing cyanobacterial concentration, pH, total nitrogen, dissolved oxygen, turbidity, and chlorophyll-a. Regarding cyanobacteria, two species were dominant: Oscillatoria sp. and Microcystis sp. while seven were abundant: Merismopedia sp., Cylindrospermopsis sp., Pseudanabaena sp., Anabaena sp., Aphanizomenon sp., Aphanocapsa sp. e Dolichospermum sp. The presence of potentially toxic cyanobacteria and the worsening of water quality make frequent monitoring and the development of alert systems essential for the management of water resources, both from an environmental and public health point of view.

ABSTRACT

On September 15, 2020, a patient came in to Peking Union Medical College Hospital after suffering from chest pain, rash and lymphadenopathy for more than 2 years, and expericing general pain for 8 months. He also lost 15 kilograms of weight in the past half a year. For chest pain, cough, and expectoration symptons, cephalosporins and symptomatic treatments were empirically used and the symptoms were relieved. However, drug-induced rash occurred. After anti-allergic treatments, the rash was relieved but the swelling of cervical lymph nodes was not relieved. Diagnostic antituberculosis therapy was employed after biopsy result showed multifocal granulomatous inflammation. The therapy reduced the lymph nodes, but the condition repeated, and the whole body pain appeared. Further examinations showed that lung cancer in the right upper lobe was accompanied by obstructive pneumonia and whole body metastasis to multiple sites. The patient had no history of immunosuppression, and the γ interferon antibody was strongly positive, adult-onset immunodeficiency syndrome was considered. In order to confirm the diagnosis, bone and tissue biopsy were necessary, but the patient failed to cooperate. The histopathological examination results of the right cervical lymph node and the left iliac bone biopsy were weakly acid fast staining positive, Nocardia infection was suspected. The clinical symptoms improved after using sulfanilamide and imipenem empirical treatments against Nocardia. The content of cyanobacteria marneffei, which was detected by metagenomic next generation sequencing, was low. More laboratory pathogenic examinations were actively confirmed for this rare pathogen. The antifungal treatment (amphotericin B plus itraconazole) was used after successfully cultivating the cyanobacteria marneffei pathogen. After two months, the body nodules and masses disappeared and the skin wounds healed. It is of great significance to the diagnosis and treatment of patients to obtain qualified samples, maintain timely communication between the laboratory and the clinic, and construct reasonable interpretations of the results of metagenomic next generation sequencing technology.

ABSTRACT

Cyanobacteria are important photosynthetic autotrophic microorganisms and are considered as one of the most promising microbial chassises for photosynthetic cell factories. Glycogen is the most important natural carbon sink of cyanobacteria, playing important roles in regulating its intracellular carbon distributions. In order to optimize the performances of cyanobacterial photosynthetic cell factories and drive more photosynthetic carbon flow toward the synthesis of desired metabolites, many strategies and approaches have been developed to manipulate the glycogen metabolism in cyanobacteria. However, the disturbances on glycogen metabolism usually cause complex effects on the physiology and metabolism of cyanobacterial cells. Moreover, the effects on synthesis efficiencies of different photosynthetic cell factories usually differ. In this manuscript, we summarized the recent progress on engineering cyanobacterial glycogen metabolism, analyzed and compared the physiological and metabolism effects caused by engineering glycogen metabolism in different cyanobacteria species, and prospected the future trends of this strategy on optimizing cyanobacterial photosynthetic cell factories.

Abstract Zooplankton are widely recognised as being regulated primarily by predators and food availability. In reservoirs, the quantity and quality of food resources are generally affected by the characteristics of the water, which in turn are controlled by the flow pulse generated by operation of the dams. In this study, we investigated the relationship between zooplankton, water quality and food availability (phytoplankton) in eight hydroelectric reservoirs located in Brazil. Samples were collected during the rainy and dry periods between 2008 and 2009. In general, the reservoirs exhibited mesotrophic conditions and Cyanobacteria were the predominant phytoplankton. The results showed that the rotifers Kellicottia bostoniensis, Hexarthra mira, Keratella spp., and Polyarthra vulgaris were present, indicating nutrient-rich environments. In addition, the copepod Thermocyclops decipiens occurred in eutrophic environments. In contrast, the cladoceran Daphnia gessneri and copepod Notodiaptomus henseni were considered indicators of more desirable water quality, owing to their relationship with waters with lower levels of nutrients and suspended solids. The results support the use of these organisms as a useful tool for understanding changes in water quality and in the ecosystem processes involved.


Resumo O zooplâncton é amplamente reconhecido como sendo regulado principalmente por predadores e pela disponibilidade de alimento. Em reservatórios, a quantidade e a qualidade de recursos alimentares são afetadas pelas características da água que, por sua vez, são controladas pelo pulso de fluxo gerado pela operação das barragens. Neste estudo, investigamos a relação entre o zooplâncton, qualidade d'água e a disponibilidade de alimento (fitoplâncton) em oito reservatórios hidrelétricos localizados no Brasil. Amostras foram coletadas durante os períodos chuvoso e seco, entre os anos de 2008 e 2009. Em geral, os reservatórios exibiram condições mesotróficas e Cyanobacteria foi o fitoplâncton predominante. Os resultados mostraram que os rotíferos Kellicottia bostoniensis, Hexarthra mira, Keratella spp. e Polyarthra vulgaris foram indicadores de ambientes ricos em nutrientes. Além disso, o copépode Thermocyclops decipiens ocorreu em ambientes eutróficos. Por outro lado, o cladócero Daphnia gessneri e o copépode Notodiaptomus henseni foram considerados indicadores de melhor qualidade da água, devido a sua relação com águas com baixos níveis de nutrientes e sólidos em suspensão. Os resultados suportam o uso desses organismos como uma ferramenta útil para o entendimento das mudanças na qualidade d'água e nos processos ecossistêmicos envolvidos.

Abstract The cyanobacterial exopolysaccharides (EPSs) are considered as one of the important group of biopolymers having significant ecological, industrial, and biotechnological importance. Cyanobacteria are regarded as a very abundant source of structurally diverse, high molecular weight polysaccharides having variable composition and roles according to the organisms and the environmental conditions in which they are produced. Due to their structural complexity, versatility and valuable biological properties, they are now emerging as high-value compounds. They are possessing exceptional properties and thus are being widely explored for various applications like in food and pharmaceutical industries, in bioremediation for removal of heavy metals, for soil conditioning, as biopolymers, bioadhesives, and bioflocculants. However, poor understanding of their complex structural properties, lack of concrete information regarding the genes encoding the proteins involved in the EPS biosynthetic pathways, their process of production and about the associated factors controlling their structural stability, strongly limits their commercialization and applications in the various fields of biotechnology. Owing to the above context, the present review is aimed to organize the available information on applications of cyanobacterial EPSs in the field of biotechnology and to identify the research gaps for improved industrial utilization and commercialization of these biomaterials.

Lactate is an important industrial chemical and widely used in various industries. In recent years, with the increasing demand for polylactic acid (PLA), the demand for lactate raw materials is also increasing. The contradiction between the high cost and the market demand caused by the heterotrophic production of lactate attracts researchers to seek other favorable solutions. The production of lactate from photosynthetic carbon fixation by cyanobacteria is a potential new raw material supply strategy. Based on the photosynthetic autotrophic cell factory, it can directly produce high optical purity lactate from carbon dioxide on a single platform driven by solar energy. The raw materials are cheap and easy to obtain, the process is simple and controllable, the products are clear and easy to separate, and the double effects of energy saving and emission reduction and production of high value-added products are achieved at the same time, which has important research and application value. This paper reviews the development history of cyanobacteria carbon sequestration to produce lactate, summarizes its research progress and encounters technical difficulties from the aspects of metabolic basis, metabolic engineering strategy, metabolic kinetics analysis and technical application, and prospects the future of this technology.

ABSTRACT

Cyanobacteria-mediated silver nanoparticles synthesis approach has proven to be more efficient and eco-friendlyin achieving biomedical applications compared to physical and chemical prototypes. In the present work, the silvernanoparticles were successfully synthesized by cell-free extract of freshwater cyanobacteria, i.e., Chroococcus turgidusand Characium typicum. The cyanobacterial silver nanoparticles (CSNPs) were characterized by UV-Vis spectroscopy,scanning electron microscopy (SEM), transmission electron microscopy (TEM), and Fourier transform infrared (FTIR)analysis and were further tested for antibacterial and cytotoxic efficiency. The synthesis of CSNPs was confirmed throughvisible color change and shift of peaks at 430–445 nm by the UV-Vis spectroscopy. The size of CSNPs was between22 and 34 nm and oval-shaped which were confirmed by SEM and TEM analyses. The FTIR spectra showed a newpeak at the range of 3,400–3,460 cm−1 compared to control, confirming the reduction of silver nitrate. Furthermore, theantibacterial activity of CSNPs showed highest zone of inhibition with 6.9, 4.0, 2.0, and 3.0 mm against Salmonellaparatyphi, Escherichia coli, Klebsiella pneumonia, and Staphylococus aureus, respectively, whereas in vitro cytotoxicactivity of C. typicum and C. turgidus silver nanoparticles showed remarkable IC50 values with 43.3 and 40.9 ug/mlagainst MCF-7 breast cancer cell line and 20.8 and 55.7 ug/ml against HepG2 cancer cell, simultaneously

Aim: Antarctic cyanobacteria may represent a potential resource of new and unique compounds with interesting capabilities. Profiling of fatty acids in Antarctic cyanobacteria can provide an overview of potential fatty acids present in them, that can be utilised in future applicationsMethodology: In total, 4 cyanobacteria previously isolated from Antarctic polar ice was used in this study. Molecular identification using 16S rRNA gene was used to ascertain their identities as Leptolyngbya spp. and their fatty acid profiles were determined using GCMS. Results: Morphologically, these cyanobacteria were found similar to Leptolyngbya sp. Analysis of 16S rRNA gene sequences amplified further confirmed their identity and were designated as Leptolyngbya sp. A, B, D and E. Following fatty acid analysis using GCMS, it was determined that unsaturated fatty acids predominated the fatty acid profiles for Leptolyngbya sp. A, B and D, while saturated fatty acid was found dominant in Leptolyngbya sp. E. Leptolyngbya sp. D contained almost 100% of linoleic acid, whilst Leptolyngbya sp. A and Leptolyngbya sp. B contained 59.35% and 83.33% of linoleic acid, respectively. Besides linoleic acid, palmitoleic acid (18.45%), oleic acid (19.45%) and lauric acid (2.74%) were also present in Leptolyngbya sp. A. As for Leptolyngbya sp. B, other than linoleic acid, only oleic acid (16.67%) was detected. Interpretation: Findings from this study demonstrate that the Antarctic Leptolyngbya spp. A, B and D identified in this study possess high content of unsaturated fatty acids, while only saturated fatty acid was present in Leptolyngbya sp. E. Fatty acid profiles revealed the potential of these Antarctic Leptolyngbya species to be further exploited for other applications.

ABSTRACT

Los microorganismos fijadores de nitrógeno de vida libre, abarcan una gama morfológica que va desde los organismos unicelulares como las bacterias y algunas cianobacterias, hasta multicelulares, filamentosas, por ello es importante conocer cómo se comportan y se puede saber haciendo una curva de crecimiento microbiano. Para este estudio se prepararon 4 fotobioreactores de columna burbujeada con inoculo de Fischerella TB22, se pusieron en aireación constante con 12 horas luz y 12 horas obscuridad durante 40 días con diferentes tratamientos de ajuste de volumen del medio de cultivo y ajuste del pH. El objetivo de este trabajo fue evaluar el crecimiento en biomasa por peso seco, densidad óptica, pH y amonio de Fischerella sp. en medio de cultivo BG110 durante 12 días. Las variables que se midieron de la curva de crecimiento de las cianobacterias, siguieron el patrón de una curva típica de crecimiento microbiano. (AU)


Free-living nitrogen-fixing microorganisms cover a morphological range that goes from unicellular organisms such as bacteria and some cyanobacteria, to multicellular, filamentous, therefore it is important to know how they behave and can be known by makinga microbial growth curve. For this study, 4 bubbled column photobioreactors with Fischerella TB22inoculum were prepared, they were placed in constant aeration with 12 hours of light and 12 hours of darkness for 40 days with different treatments of volumeadjustment of the culture medium and pH adjustment. The objective of this work was to evaluate the biomass growth by dry weight, optical density, pH, and ammonia of Fischerella sp. in the BG110 culture medium for 12 days. The variables that were measured from the growth curve of cyanobacteria followed the pattern of a typical microbial growth curve. (AU)

ABSTRACT

Resumen El presente trabajo reporta por primera vez para el Perú a la cianobacteria bentónica Lemmermanniella uliginosa, identificada en muestras de perifiton y sedimentos bentónicos procedentes del humedal de Caucato en el distrito de San Clemente, departamento de Ica. Además, se registra por primera vez al género Lemmermanniella para el país. Se discuten aspectos morfo-taxonómicos de la especie comparándola con poblaciones reportadas para otras localidades en zonas tropicales.


Abstract This work presents the first record of Lemmermmanniella uliginosa from Peru based on periphyton and sediment samples from Caucato wetland (San Clemente district, Ica department). Furthermore, the genus Lemmermmanniella is recorded for the first time for Peru. Morpho-taxonomic comparison with other populations reported in tropical regions is discussed.

ABSTRACT

Abstract Nostoc is a genus of filamentous cyanobacteria with biotechnological applications in human nutrition, biomedicine, biofertilization and commercial production of biofuels. However, the low growth rate in liquid medium due to its periphytic nature and its tendency to form biofilms, limits its large-scale production. Therefore, the aim of this study was to evaluate the biomass production of Nostoc muscorum in a modified hydroponic system. Cultures of N. muscorum were made by triplicate, in a hydroponic system under semicontrolled conditions of temperature (29 ± 13 °C), light intensity (32 ± 54 µmol/m2/s) and photoperiod (12 hours), for a total of 23 days inside a greenhouse. Temperature, pH, conductivity and dry biomass production were monitored on alternating days. The results showed that the maximum dry biomass production was 0.2276 ± 0.0114 g/m2/day, and the average productivity was 0.4149 ± 0.0207 g/m2/day. The maximum biomass production of N. muscorum was achieved on day thirteen with 0.3185 ± 0.0159 g/m2/day. The correlation statistical analysis of environmental variables did not show significant differences; thus, temperature, pH and electrical conductivity did not affect the biomass production of N. muscorum. Consequently, the algal growth was influenced by the species physiology only. The support used in the hydroponic system allowed the adhesion and development of the algae mucilaginous layer without requiring drying periods as in conventional crops. The hydroponic system provided a continuous flow of nutrients hat could prevent the attack of opportunistic bacteria and fungi, generating a high growth rate of N. muscorum. The hydroponic system represents a viable alternative for the production of N. muscorum biomass under greenhouse conditions at large scale.

Sin embargo, su baja tasa de crecimiento en medio líquido por su naturaleza perifítica y su tendencia a formar biofilms, limita su producción a gran escala. Por lo tanto, el objetivo de este estudio fue analizar la producción de biomasa de Nostoc muscorum en un sistema hidropónico modificado. Para ello, se realizaron cultivos de N. muscorum por triplicado, en un sistema hidropónico bajo condiciones semicontroladas de temperatura (29 ± 13°C), intensidad lumínica (32 ± 54 µmol/m2/s) y fotoperiodo (12 horas), durante 23 días en un invernadero. La temperatura, el pH, la conductividad eléctrica y la producción de biomasa seca, fueron monitoreados en días alternados. Los resultados arrojaron que la producción máxima de biomasa seca fue de 0.2276 ± 0.0114 g/m2/día, y la productividad promedio fue de 0.4149 ± 0.0207 g/m2/día. A su vez, la producción máxima de biomasa de N. muscorum se obtuvo el día trece con 0.3185 ± 0.0159 g/m2/día. El análisis estadístico de correlación de variables ambientales no arrojó diferencias significativas, por lo que la temperatura, el pH y la conductividad eléctrica no afectaron la producción de biomasa de N. muscorum. Consecuentemente, el crecimiento algal fue influenciado por la fisiología de la especie. El soporte empleado en el sistema hidropónico permitió la adherencia y el desarrollo de la capa mucilaginosa de la cianobacteria sin requerir períodos de desecación como en los cultivos convencionales. El sistema hidropónico proporcionó un flujo continuo de nutrientes que podría prevenir el ataque de bacterias y hongos oportunistas, generando una alta tasa de crecimiento. De este modo, este sistema hidropónico representa una alternativa viable para la producción de biomasa de N. muscorum en condiciones de invernadero a gran escala.

ABSTRACT

Anabaena sp. are the dominant cyanobacterial species on terracotta monuments of Bishnupur which exposed to high solar radiation, ultraviolet and in a desiccated condition in most part of the year. In the present study three Anabaena species were isolated from crust samples and its antibacterial activities were evaluated against pathogenic bacteria Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Salmonella typhimurium, Escherichia coli. We observed good antibacterial activity in ethyl acetate and ethanol extract of Anabaena sp. (VBCCA 052002) which having highest antibacterial activity against Staphylococcus aureus, Salmonella typhimurium, Staphylococcus aureus and E. coli respectively. We have validated the antibacterial assay by using resazurin based antimicrobial assay in microtiter plates and calculated the MIC value of ethyl acetate extract of Anabaena sp. (VBCCA 052002) which is found to be 100 µg/ml against Staphylococcus aureus and 150 µg/ml against Salmonella typhimurium.

Abstract The objective of this study was to characterize the limnological, microystin and phytoplankton community of five tropical eutrophic reservoirs located in the Brazilian northeastern semi-arid region, used for domestic use at the time of extreme drought and reduction of water volume. The study was conducted in July and August 2015, and an integrated sample of the water column was collected at three points near the dam in each reservoir. Analysis of limnological parameters, identification and quantification of phytoplankton, with emphasis on cyanobacteria were performed, as well as detection of microcystin by means of immunoassay (ELISA). The reservoirs presented ~ 90% water volume reduction. High turbidity and concentrations of nitrogen and phosphorus, as well as high cyanobacterial densities, revealed an increase in the eutrophic state for hypereutrophy. The total biovolume of phytoplankton and cyanobacterial density is high, plus an average increase in relation to previous studies of 350% and 150%, respectively. The density of cyanobacteria and microcystin concentration presented values ​​above acceptable levels for drinking water according to Brazilian legislation. A phytoplankton community was represented by 17 functional groups, including potentially toxic cyanobacteria species such as Planktothrix agardhii (S1), Microcystis aeruginosa (M), Anabaena planktonica e Anabaena spp. (H1), Cylindrospermopsis raciborskii (Sn). Our results confirm that conditions of extreme drought and reduction of the volume of the reservoirs influence the composition, biovolume of phytoplankton and water quality, but not the increase of total microcystin in the analysed, although above 1μg-1 registered a significant decrease of water quality in used for human consumption.


Resumo O objetivo deste trabalho foi caracterizar a comunidade limnológica, microistalina e fitoplanctônica de cinco reservatórios eutróficos tropicais localizados no semi-árido nordestino brasileiro, utilizados para uso doméstico no período de seca extrema e redução do volume de água. O estudo foi realizado em julho e agosto de 2015, e uma amostra integrada da coluna de água foi coletada em três pontos próximos à barragem em cada reservatório. Análises de parâmetros limnológicos, identificação e quantificação do fitoplâncton, com ênfase em cianobactérias, foram realizadas, assim como a detecção de microcistina por meio de imunoensaio (ELISA). Os reservatórios apresentaram ~ 90% de redução do volume de água. A alta turbidez e as concentrações de nitrogênio e fósforo, bem como as altas densidades de cianobactérias, revelaram um aumento no estado eutrófico da hipereutrofia. O biovolume total de fitoplâncton e densidade de cianobactérias é alto, além de um aumento médio em relação a estudos anteriores de 350% e 150%, respectivamente. A densidade de cianobactérias e a concentração de microcistina apresentaram valores acima dos níveis aceitáveis ​​para água de consumo, de acordo com a legislação brasileira. Uma comunidade fitoplanctônica foi representada por 17 grupos funcionais, incluindo espécies de cianobactérias potencialmente tóxicas, como Planktothrix agardhii (S1), Microcystis aeruginosa (M), Anabaena planktonica e Anabaena spp. (H1), Cylindrospermopsis raciborskii (Sn). Nossos resultados confirmam que condições de seca extrema e redução do volume dos reservatórios influenciam a composição, o biovolume de fitoplâncton e a qualidade da água, mas não o aumento do total de microcistina no analisado, embora acima de 1μg-1 tenha registrado uma diminuição significativa da qualidade da água usado para consumo humano.

ABSTRACT

O presente trabalho tem como objetivo estimar o volume de esgoto gerado no RS em Demanda Bioquímica de Oxigênio (DBO kg/dia), e o volume de esgoto (DBO kg/dia) que chega até as estações de tratamento de esgoto ETEs; diagnosticar a eficiência das estações de tratamento de esgotoem relação ao percentual de remoção de (DBO kg/dia); correlacionando com a presença de cianobactérias próximas aos pontos de captação de água superficial para consumo humano; analisar se há inativação ou remoção de protozoários no lançamento do efluente ao retornar ao manancial. Como resultado obteve-se o volume total de matéria orgânica lançada nos corpos hídricos por dia estimado no Rio Grande do Sul é 511.769,00 DBO kg/dia no RS. As Estações de Tratamento de Esgoto captam 390.285,45 DBO kg/dia apresentaram resultados de remoção de 145.500,50 DBO kg/dia e de 22.963,21 DBO kg/dia residual; O volume ocioso das ETEs fica em torno de 5.049,89 L/s. O processo de eliminar ou inativar grande quantidade de protozoários presentes no afluente se dá através de etapas terciárias avançadas nas ETEs. Não encontramos no RS no ano 2019 ETEs com esses tipos avançados de tratamento. Sugere-se então que haja um melhor aproveitamento destas estações de tratamento no que se refere à quantidade e qualidade de volume de esgoto tratado e que sejam ampliados estudos para viabilidade de projetos de execução de novas ETEs para os demais municípios que despejam seus esgotos in natura nos mananciais impactando-os.


This work aims to estimate the volume of sewage generated in RS in Biochemical Oxygen Demand (BOD kg/day), and the volume of sewage (BOD kg/day) that reaches the sewage treatment stations (STS)s; diagnose the efficiency of sewage treatment station in relation to the percentage of removal (BOD kg/day); correlating with the presence of cyanobacteria close to the surface water intake points for human consumption; analyze if there is inactivation or removal of protozoa in the discharge of the effluent when returning to the source. As a result, the total volume of organic matter released into water bodies per day estimated in Rio Grande do Sul is 511,769.00 BOD kg/day in RS. The Sewage Treatment Plants capture 390,285.45 BOD kg/day showed removal results of 145,500.50 BOD kg/day and 22,963.21 BOD kg/day residual; The idle volume of ETEs is around 5,049.89 L/s. The process of eliminating or inactivating a large amount of protozoa present in the tributary takes place through advanced tertiary stages in the STSs. We did not find in RS in the year 2019 STSs with these advanced types of treatment. It is therefore suggested that there is a better use of these treatment plants with regard to the quantity and quality of the volume of treated sewage and that studies for the feasibility of projects for the execution of new STSs be expanded to the other municipalities that dump their raw sewage in water sources impacting them.

ABSTRACT Almost half of the Brazilian population has no access to sewage collection and treatment. Untreated effluents discharged in waters of reservoirs for human supply favor the flowering of cyanobacteria - and these microorganisms produce toxins, such as saxitoxin, which is a very potent neurotoxin present in reservoirs in the Northeast region. A recent study confirmed that chronic ingestion of neurotoxin-infected water associated with Zika virus infection could lead to a microcephaly-like outcome in pregnant mice. Cyanobacteria benefit from hot weather and organic matter in water, a condition that has been intensified by climate change, according to our previous studies. Considering the new findings, we emphasize that zika arbovirus is widespread and worsened when associated with climate change, especially in middle- or low-income countries with low levels of sanitation coverage.


RESUMO No Brasil, quase metade da população não tem acesso a coleta e tratamento de esgotos. Os efluentes não tratados, devido a contaminação das águas de reservatórios para abastecimento humano, geram florações de cianobactérias - e esses microrganismos produzem toxinas, como a saxitoxina, uma neurotoxina bastante potente e presente nos reservatórios da região Nordeste. Estudo recente confirmou que a ingestão crônica de água contaminada com neurotoxinas associada a infeção pelo zika vírus em camundongos prenhes poderia levar a desfecho semelhante a microcefalia. Cianobactérias são beneficiadas por tempo quente e matéria orgânica na água, e essas condições estão sendo intensificadas pelas mudanças climáticas, segundo nossos estudos anteriores. Aqui, ressaltamos que, à luz dos novos achados, a arbovirose do zika, associada às mudanças do clima, se amplifica e se agrava, sobretudo em países de média ou baixa renda, com baixos níveis de cobertura de saneamento.
